Complex diseases such as heart disease, cancer, diabetes and chronic obstructive pulmonary disease (COPD) are the leading causes of death worldwide. Due to the multigenic risk, lifestyle, and environmental factors our understanding of the pathobiology of these diseases remains limited, which in turn limits the development of effective therapies. Our division at Brigham and Women’s Hospital is one of the leading laboratories investigating chronic respiratory diseases with millions of biosamples collected over decades supporting our efforts to uncover the risk factors and related biological mechanisms. In this talk I will present the general problem of complex diseases, „omics” research, and our recent results in modelling the disease mechanisms of top risk genes using dynamic network models. These models offer systems-level mechanistic insights into how genetic risk translates into disease phenotypes. I will also discuss our computational and experimental validation methods of our network models, with insights into potential new therapies.
